An assassin, working for the Illuminati, has captured four cardinals, and murders each, painfully. Robert and Vittoria also are searching for a new very destructive weapon that could kill millions. Despite his notorious relationship with the Roman Catholic church, Harvard symbologist Robert Langdon is once again called upon to decipher the clues to a catastrophic conspiracy.

Harvard symbologist Robert Langdon must join forces with scientist Vittoria Vetra in a frantic race to save the Vatican from a powerful bomb which threatens to destroy it and the cardinals who have gathered within the Sistine Chapel to elect the next Pope. Their journey takes them across Rome and into cathedrals, secret archives and the Illuminati lair. Along the way, they uncover many secrets, including who is responsible for the threat to the Vatican. In the end, they save the Vatican and the cardinals from harm. Langdon is summoned to Switzerland when it is discovered that an Illuminati brand was used to mark a dead man's chest. Vittoria and her father have created a sample of antimatter, a highly unstable material, which has been stolen.

The novel introduces the character Robert Langdon , who recurs as the protagonist of Brown's subsequent novels. Ancient history , architecture, and symbology are also heavily referenced throughout the book. A film adaptation was released on May 15, The book contains several ambigrams created by real-life typographer John Langdon. The book also contains ambigrams of the words Earth , Air , Fire , and Water , which has served to bring the art of ambigrams to public attention by virtue of the popularity of the book. CERN director Maximilian Kohler discovers one of the facility's top physicists, Leonardo Vetra, murdered, his chest branded with an ambigram of the word " Illuminati ".

It boasted a fascinating protagonist, a Harvard professor of religious iconography named Robert Langdon, and compelling conspiracy theories. Brown, it seemed, had come out of nowhere. It's major themes are religion versus science, skepticism versus faith, and the hold that powerful people and institutions have over the people they supposedly serve. It introduced the general public to an ages-old secret society, and was a unique entry into the world of conspiracy theory mysteries. Packing the novel with sinister figures worthy of a Medici, Brown sets an explosive pace through a Michelin-perfect Rome. The book received its share of criticism, mainly for its historical inaccuracies presented as fact, a criticism that would carry over into "The Da Vinci Code," which played even more fast and loose with history and religion. Some Catholics took offense at "Angels and Demons," and with its subsequent sequels, stating that the book is nothing but a smear campaign of their beliefs.
